A methods paragraph
is a prescription
"The PCR mix contained 10 U of LA Taq, 10x buffer II, dNTP Mixture (200 nM each), 4.8 ng DNA Template…" — six reagents in one sentence.
This course unfolds the methods section of Wei 2023 into a list and picks up thirty reagents one at a time: what each one is, why that concentration, and what breaks if you leave it out. Only four are enzymes; everything else is there to help those four.
Python runs right inside this page. There is nothing to install.
How you'll learn
It starts by unfolding the methods prose into a table. Once unfolded, the questions appear by themselves.
LA Taq, T7, DNase, E-PAP. Of thirty reagents four do the work; the rest are there to help.
For each reagent we write down what happens without it — the fastest way to learn one.
0.72 M, 1.875 mM, 0.5 U/uL, 4.8 ng. Anything not in the paper is marked as an assumption.
